Safranin (also called Safranin O or basic red 2 ), is a biological contrast dye used in Gram staining to provide a more intense violet color to Gram+ bacteria and to stain G- bacteria pink; in histology and cytology. Safranin is used as a contrast liquid in some staining protocols, staining the cell nucleus red. Also to detect cartilage, mucin and mast cell granules.
Safranin is a dimethyl safranin . There is also a trimethyl safranin , which has a methyl group added to the ortho- position of the lower ring. Both compounds behave essentially identically in biological staining applications, and many safranin manufacturers do not distinguish between the two. Commercial preparations of safranin frequently have a mixture of both.
Safranin is also used as a redox indicator in analytical chemistry.
